CML is caused due to reciprocal translocation between Chr 9 and Chr 22 leading to formation of Philadelphia Chromosome'.
Diagnosis of CML can be done easily on simple Peripheral blood smear and confirmed with Bone marrow examination and FISH testing for Philadelphia Chromosome.
